A method for pretreatment of a biological sample for examining the distribution of a drug in a biological sample or quantifying the drug in the biological sample using matrix-assisted laser desorption / ionization (MALDI). A step of forming a matrix crystallite on the surface of the biological sample containing the drug by depositing the matrix on the biological sample, and further dispensing a matrix solution on the biological sample; A method for preparing a sample for mass spectrometry, which comprises a step of growing a matrix crystal on the microcrystal by spraying.
