abstract | To measure the distribution of ammonia in a biological sample. A matrix which is DHB or DHAP is attached to a sample surface by vapor deposition under an atmospheric pressure or a low vacuum atmosphere (S2), and the sample is subjected to mass analysis by an imaging mass spectrometer using an atmospheric pressure MALDI ion source. (S3). From the mass spectrum obtained for each minute region, the signal intensity of the ammonium ion adduct and the potassium ion adduct of lipid in the sample are obtained, the signal intensity ratio is calculated (S4), and the two-dimensional distribution of the signal intensity ratio A heat map is displayed to create an imaging image (S5). By performing sample preparation by low vacuum vapor deposition and ionization at atmospheric pressure, it is possible to suppress volatilization and decomposition of ammonia in the sample. In addition, by using an ammonium ion adduct of lipid that is difficult to move in living cells as an analysis target, it is possible to grasp an accurate distribution of ammonia.
