It is intended to provide a medium, by which the problem of charge-up can be easily solved and the practical morphology, etc. of a sample can be observed under an SEM, a TEM, etc., a preparation, a method of constructing the preparation, a method of observing the preparation, a sample cell and an electron microscope. To achieve the object as described above, use is made of a liquid medium to be used in an electron microscope for imparting electrical conductivity which contains an ionic medium as the essential component and is used in impregnating a whole sample for an SEM or a TEM therewith or coating the observation face therewith to impart electrical conductivity at least to the observation face. Since the electrical charge stored on the sample surface can be released in the above method merely by impregnating or coating the sample with the ionic liquid, the problem of charge up can be easily solved. Moreover, the ionic liquid would never evaporate when the sample having been impregnated or coated therewith is allowed to stand in vacuo, which makes it possible to observe a biological sample while sustaining its original shape. |
