titleOfInvention Immobilized enzyme, and production of optically active indoline-2-carboxylic acid therewith
abstract PURPOSE:To produce optically active indoline-2-carboxylic acid efficiently and industrially, by subjecting an (R,S)-indoline-2-carboxylic acid ester to asymmetric hydrolysis with a specific immobilized enzyme. CONSTITUTION:An immobilized enzyme (immobilized microorganism may be used) having stereoselective esterase activity, supported on a hydrophobic carrier, is used. Namely, (i) an (R,S)-indoline-2-carboxylic acid ester shown by the formula I (R is 2-8C aliphatic hydrocarbon) is loaded into a column packed with the immobilized enzyme, (ii) a buffer solution is made to flow into it to carry out an asymmetric hydrolysis reaction, prepared hydrophilic optically active indoline-2-carboxylic acid shown by the formula II is eluted with a buffer solution, and recovered. Then, (iii) an organic solvent with low polarity (e.g., benzene, or hexane) is made to flow into it, an adsorbed hydrophobic indoline-2- carboxylic acid ester shown by the formula III is eluted and recovered.
