abstract Methods for isolating protein subpopulations involved in protein-protein interactions The present invention provides methods for isolation and identification of proteins involved in protein-protein interactions in complex mixtures. The method uses a chemically reactive support matrix to isolate proteins that are in turn non-covalently bound to other proteins. The support matrix is isolated and the non-covalent protein is released for subsequent analysis. Since the protein is susceptible to chemical manipulation at the binding and release stages, identification of non-covalently bound proteins is a specific classification of interacting proteins, such as information on calcium-dependent or substrate-dependent protein interactions. give. This makes it possible to select a sub-population of proteins from the complex mixture based on defined interaction criteria. Unlike two-hybrid systems or phage display, where only proteins bound to single-byte proteins can be detected at one time, the method allows simultaneous screening of all proteomes.
