The present invention regards a method of amplifying GC-rich molecules, comprising: providing DNA molecules; subjecting the DNA molecules to heat conditions to denature DNA molecules with low GC content but not to denature DNA molecules with high GC content; ligating an adaptor onto the end of the DNA molecules with high GC content to produce adaptor-ligated molecules; and amplifying the adaptor-ligated molecules with high GC content. |
