DNA encoding a 3-phosphoglycerate dehydrogenase (PGD) exhibiting reduced sensitivity to inhibition by serine compared to the wild-type PGD, in other words DNA encodes a PGD which exhibits at least a certain level of enzymatic activity which can be used for biosynthesis, and which retains this activity in the presence of serine levels higher than the PGD of wild phenotype (unmodified). Also described are: a) a PGD containing the amino acid sequence of the modified DNA described above; b) expression vectors comprising the modified DNA and a regulatory DNA positioned and oriented so as to express the modified DNA in a host expression system; c) cells comprising such expression vectors; and d) methods of producing serine or a serine product by culturing such cells. |
