abstract | This invention relates to a method of detecting two or more target polynucleotide sequences comprising using a probe oligonucleotide A0 annealing to the target, whereafter the 3' end is pyrophosphorolysed and the remaining probe is circularised or ligated to another probe using a splint oligonucleotide, and a padlock probe X0, which is circularised against the target and not pyrophosphorolysed, and then a signal from (amplicons of) the ligated probes is detected to indicate presence of the target. Further disclosed are kits comprising the probes and enzymes as well as buffer and a source of pyrophosphate ions. |
