abstract | Disclosed is a single-base continuous extension flow-type targeted sequencing method. The method uses a single-stranded nucleotide fragment containing a complementary sequence of a nucleic acid fragment to be tested, a primer, microspheres, nucleic acid polymerase, a fluorescently labeled ddNTP, a fluorescently labeled dNTP in which ribose 3'-OH is protected, a dNTP in which ribose 3'-OH is protected, a ribose 3'-OH deprotecting agent, and so on, and, by means of the single base extension of the primer, a series of sequencing microspheres suitable for using a flow cytometer to detect a nucleic acid base sequence is obtained. Said sequencing microspheres further perform detection by using a flow cytometer, and the base sequence of said nucleic acid fragment can be obtained. |
