abstract | Provided are a method for detecting and a method for quantifying oligonucleotides, the methods exhibiting excellent specificity and quantification compared to conventional signal amplification (PALSAR) measurement methods. In the present invention: a target oligonucleotide to be detected is hybridized with a complementary nucleic acid probe (complementary sequence-5' of 3'-target sequence), or a target oligonucleotide to be detected has a given base (poly A or the like) added thereto and is then hybridized with a complementary nucleic acid probe (complementary sequence-5' of 3'-target sequence + given base); a single-strand-specific nuclease is used to decompose and remove incomplete hybridization product; and the nucleic acid probe included in the remaining complete hybridization product is measured using a PALSAR method. |
