abstract | The invention relates to molecular biology, biotechnology, medicine and can be used for the production of human growth hormone. A plasmid DNA for the synthesis of human growth hormone in Escherichia coli cells is proposed, represented by the vector pET-21a (+), the sequence from the T7 promoter by lac operator is modified and characterized by SEQ ID NO: 3 containing the insert of the gene encoding a protein characterized by the amino acid sequence SEQ ID NO: 1 optimized for codon composition for expression in Escherichia coli cells. Also proposed is a strain producing human growth hormone, based on Escherichia coli BL21 (DE3) cells transformed with the described plasmid DNA. The use of the proposed inventions allows to achieve a significant increase in the yield of the monomeric form of somatotropin, up to 80%. |
