abstract |
Olmsted's syndrome (SO) is a rare genodermatosis. The disease is debilitating and progressive keratoderma and finger self-amputation can prevent patients from grasping objects and walking, and confining them to a wheelchair. New therapeutic options are therefore crucial and are expected through a better understanding of the mechanisms of the disease. The inventors have demonstrated abnormal activation of the mTOR pathway in the lesional skin of patients with SO. Topical treatment with 1% Sirolimus demonstrates satisfactory tolerance and partial but real efficacy on inflammatory and hyperkeratotic lesions of the soles of the feet as observed in the treated patient. Thus, the present invention relates to a method of treating Olmsted syndrome in a subject in need thereof, comprising administering to a therapeutically active amount of an mTOR inhibitor. |
