abstract | The present invention relates to a method for producing a virus by a semi-continuous culture method and a method for mass-producing a fish virus at a high concentration by using a cell line attached to a microcarrier, which overcomes complexity and inefficiency of a conventional method (Batch culturing system, BCS) for producing viruses by utilizing continuous infectious cell lines, allow the production of virus at a high concentration without reducing the number of or killing host cell lines even with long-term semi-continuous culture, and allow fish cell lines with attachment culture characteristics to be cultured in large quantities even in confined spaces, by means of using a semi-continuous culture method (SCCCS method) using fish cell lines consecutively infected with megalocytivirus and deriving optimal culture conditions for attaching and scaling-up fish cell lines to microcarrier beads. As such, by using the present method, it is possible to efficiently and stably produce a virus by mass-producing a fish disease-causing virus a high concentration and to prepare a vaccine. |
