abstract | The present invention provides a recombinant yeast in which the shLkn-1 gene is introduced into Pichia pastoris GS115, and the amount of dissolved oxygen under conditions of an air supply of 5 to 80 v / v, a pressure of 300 to 1500 mbar, and a stirring speed of 200 to 1000 rpm. Batch fermentation step of culturing with a glycerol as a carbon source while maintaining at 10% or more, a fed-batch culture step of supplying a fermentation culture medium to the yeast, cultivation by a DO-stat method, and depletion of glycerol in the medium, and then into a carbon source The present invention relates to a method for mass production of shLkn-1 protein using a recombinant yeast comprising a semi-continuous fed-incubation step of secreting shLkn-1 into a culture medium at high concentration while culturing with methanol. According to the above method, practical use as an anticancer adjuvant is possible through mass production of shLkn-1 belonging to C6 β-chemokine having hematopoietic stem cell proliferation inhibitory function. |
