abstract | Provided is a method of enriching and/or isolating a target cell population from a plurality of cells. The plurality of cells is introduced into a separation compartment, which has an inlet and an outlet fluidly coupled to the ambience, a circumferential wall, a base and a top, whereof the base comprises the outlet. An at least essentially constant flow of a suitable liquid is introduced through the inlet of the separation compartment, and an at least essentially constant flow of liquid exits the separation compartment via the outlet. Thereby the plurality of cells is suspended in a continuous flow of liquid, which prevents the target cell population from sedimenting and from exiting the separation compartment via the outlet. In some embodiments the outlet has a porous medium with pores of a size smaller than the average cell size of cells of the target cell population. The porous medium may prevent the cell population from exiting the separation compartment via the outlet. |
