abstract Disclosed is a probe protector oligonucleotide for detecting an improved CATACLEAVE™ probe of a nucleic acid sequence from a test sample. The probe protector oligonucleotide is designed to be substantially mutually complementary to a FRET pair labeled CATACLEAVE™ oligonucleotide probe. A formation of a base pair between the probe protector oligonucleotide and a CATACLEAVE™ oligonucleotide probe that is combined to a chemical modification of a probe sequence prevents the formation of a spurious RNA:DNA hybrid during a PCR cycle, which can result in a non-specific cleavage of the oligonucleotide probe before the oligonucleotide probe is degraded by an endonuclease of an RNA sequence and before the detection of a target nucleic acid. An improved detection method is rapid and accurate and appropriate for high throughput application. Also disclosed is a kit, which is convenient, user-friendly, and reliable, for rapidly detecting a target nucleic acid sequence.
