Disclosed is a method for preparing a nuclease-resistant nucleic acid, which comprises the steps of mixing a nucleic acid composed of at least three nucleotides, a terminal deoxynucleotide transferase and a 2',4'-crosslinked nucleoside triphosphate with one another and incubating the resulting mixture. Examples of the 2',4'-crosslinked nucleoside triphosphate to be used in the method include 5-methyl-2'-O,4'-C-(methyleneoxy- methylene)uridine-5'-triphosphate, 5-methyl-2'-O,4'-C- (aminomethylene)uridine-5'-triphosphate and 5-methyl-2'-O,4'-C-(benzyloxymethylene)uridine-5'- triphosphate. According to the method, it becomes possible to impart excellent nuclease resistance to an existing functional nucleic acid afterward in a simple manner. |
