abstract | It is intended to provide a method of recovering DNA whereby a target cDNA fragment can be more surely recovered from a cDNA fragment mixture having various types of cDNA fragments. Namely, a method of recovering DNA which comprises the first PCR step wherein a mixture of cDNA fragments each having a first adaptor sequence on one end and a second adaptor sequence on the other end is subjected to PCR by using a first primer having a sequence complementary to the first adaptor sequence together with a labeling and a second primer having a sequence complementary to the second adaptor sequence; the electrophoresis step wherein the cDNA fragments having been amplified in the first PCR step are electrophoresed on a gel; and the recovery step wherein a gel segment containing the target cDNA fragment is cut out from the gel based on the results of the electrophoresis to thereby recover the cDNA fragment. |
