abstract The invention concerns a method for inserting a nucleic acid fragment in a vector, consisting in: (a) preparing a double-stranded nucleic acid molecule comprising the nucleic acid fragment to be inserted in a vector and on either side of said fragment a nucleic acid label; (b) performing the denaturation of said nucleic acid molecule and of a cleaved vector whereof the 5' and 3' ends of one of the strands are complementary to said nucleic acid labels; (c) incubating said nucleic acid molecule with the cleaved vector in conditions enabling said nucleic acid labels to be hybridised on the cleaved vector; (d) copying the cleaved vector and the nucleic acid fragment; (e) ligating the newly synthesised strands with the free ends; (f) optionally repeating steps (b) through (e).
