titleOfInvention | Method of producing and using a highly purified mineral matrix in the form of segments and granules with osteoinductive properties for bone defect replacement |
abstract | FIELD: biotechnology.SUBSTANCE: invention relates to the field of biotechnology, namely to production of highly purified mineral matrix with osteoinductive properties, intended for replacement of bone tissue defects, from biological material, which is bones tissue of mammals, and its application. Method involves preliminary treatment of said biological material, including at least one freezing cycle of said material at temperatures from -20 to -80 °C and subsequent thawing at temperatures from +5 to +37 °C. Proteins and cells are then removed in a solution of ionic or amphoteric detergent in concentration of 0.1–10 % at 60–100 °C for 1–72 hours and then washing the material from the detergent in an aqueous solution. Then material is gradually heated to 300–400 °C for 6–288 hours. That is followed by impregnation of osteoinductive factors involving impregnation of the material with an osteogenic nutrient medium containing osteoinductive factors. Matrix is used as an implant for replacement of defects of bone tissue in dentistry, orthopedics or traumatology, including defects of bone tissue, resulting from congenital pathologies, benign tumors or injuries of various geneses.EFFECT: invention enables to obtain a highly purified mineral matrix with osteoinductive properties, consisting of bone hydroxyapatite and calcium phosphate, presented in native amorphous form, and additionally containing one or more osteoinductive factors, virtually free of cell proteins, peptides, lipids and nucleic acids, having a porous structure and osteoinductive potential, is sterile and can be presented in various forms.20 cl, 5 dwg, 1 tbl, 1 ex |
