titleOfInvention DETERMINATION OF GENE ZG16 mRNK LEVEL REDUCTION AS METHOD OF LARGE INTESTING CANCER DIAGNOSTICS AND SET OF ITS REALISATION
abstract FIELD: medicine. ^ SUBSTANCE: claimed invention relates to field of medicine, in particular to oncology and molecular biology and can be used for diagnostics of large intestine cancer. According to the method initial pair of tissue samples from patients are obtained, where one of the samples is obtained from tissue supposedly affected by cancer, and the second one is obtained from histologically normal (conditionally-normal) adjacent or located at a small distance from the tumour tissue; separation and purification of RNA preparations from initial pair of samples; synthesis of single-stranded cDNA on RNA matrix with application of oligonucleotide primers; fixing rate to cDNA concentration by control gene, whose transcription level is constant in norm even in case of large intestine cancer; carrying out quantitative and semi-quantitative reaction of gene ZG16 fragment amplificxation with application of cDNA obtained at stage c), as matrix and pair of gene-specific oligonucleotide primers with sequences SEQ ID NO: 1 and 2; comparison of quantity of amplified DNA ZG16 fragment for the sample obtained from the tissue supposedly affected with cancer, with the quantity of amplified DNA fragment for the sample, obtained from normal tissue, where said quantities of amplified DNA fragment represent gene ZG16 mRNA level, decrease of ZG16 mRNA quantity being a diagnostic sign of large intestine cancer.
