Enzyme improvement and screening for genes used for biosynthesis of 1,4-butanediol in E. coli |
abstract |
One embodiment is currently active against two terminal reactions of the 1,4-butanediol biosynthetic pathway (4-hydroxybutyaldehyde-producing reaction in 4-hydroxybutyryl-CoA and 1,4-butanediol-producing reaction in 4-hydroxybutyraldehyde). an unreported enzyme butyraldehyde dehydrogenase to (bld) and butanol dehydrogenase (bdh) Clostridium Saccharoperbutylacetonicum was introduced from E. coli and found to have the ability to produce 1,4-butanediol. In particular, the activity of butyraldehyde dehydrogenase enzyme was enhanced by the direction evolution method to increase the production concentration of 1,4-butanediol more than two times. |
