Proteolytically Cleavable Fusion Proteins Including Blood Coagulation Factors |
abstract |
The present invention relates to therapeutic fusion proteins in which coagulation factors are fused to half-life enhancing polypeptides, both linked by proteolytically cleavable linker peptides. Cleavage of such linkers liberates coagulation factors from any activity-lowering steric hindrance caused by half-life enhancing polypeptides, resulting in the generation of fusion proteins with high molar inactivity when tested in coagulation-related assays. In addition, the fact that the linker is cleavable can improve the inactivation rate and / or elimination rate after proteolytic cleavage of the peptide linker compared to the corresponding therapeutic fusion protein linked by an uncleavable linker having the amino acid sequence GGGGGGV. . |
