abstract | PURPOSE:To efficiently obtain an alkaline protease useful as a cleaning assistant in mass-production, by cultivating a bacterium of the genus Bacillus transformed with a plasmid containing a specific genetic DNA. CONSTITUTION:A bacterium Bacillus licheniformis (BR) is subjected to variation treatment with nitrosoguanidine to provide a variant BR bacterium having high productivity of alkaline protease (APE). The bacterial cells of the BR are then subjected to extraction treatment by a lysozyme treating method to afford a genetic DNA, having a promoter region with ability to express the APE and consisting of a base sequence expressed by the formula. The resultant DNA is subsequently linked to a vector plasmid pBR322 to afford a recombinant plasmid, which is used to transform a bacterium of the genus Bacillus (e.g. Bacillus subtilis) by a protoplast method. The obtained transformant strain is then cultivated in a culture medium containing a carbon source and nitrogen source under aerobic condition by adjusting to a desired pH and temperature to produce and accumulate APE in the culture fluid. The resultant APE is subsequently extracted, purified and recovered. |
