abstract PURPOSE: To obtain a mutant firefly luciferase by culturing a microbial strain capable of producing mutant firefly luciferase and containing a recombinant DNA having inserted mutant firefly luciferase gene. n CONSTITUTION: A mutant firefly luciferase gene coding an amino acid sequence of a wild-type firefly luciferase wherein the valine at the 233rd site is substituted with isoleucine and the valine at the 239th site is substituted with isoleucine is introduced into a vector DNA to synthesize a recombinant DNA. A microorganism belonging to genus Escherichia is transformed with the recombinant DNA and the transformant is cultured in a medium.
