abstract | PURPOSE: A nucleic acid derivative for antitumor agents, etc., composed of a polymer having stable high-order structure formed by substituting part of purine rings or pyrimidine rings of structural units with sulfur atoms and crosslinking to form disulfide bond of these sulfur atoms. n CONSTITUTION: A nucleic acid polymer containing cytidylic acid is dissolved in a mixed solvent of water with pyridine, introduced into a reactor together with liquefied hydrogen sulfide and reacted. Then the reactor is cooled down to remove the unreacted hydrogen sulfide. The resultant product is dissolved in a 50mM common salt-containing tris buffer solution and dialyzed to form a transparent liquid, followed by freeze-drying to substitute NH 2 group in the pyrimidine ring of the cytidylic acid in the molecule. Then this compound together with an equivalent amount of inosinic acid-containing nucleic acid polymer is dissolved in a buffer solution, kept warm at 68°C for 10min and 0.1N iodo-solution is added thereinto the effect reaction and obtain the aimed nucleic acid derivative having disulfide bonds. n COPYRIGHT: (C)1987,JPO&Japio |
