titleOfInvention Method of purifying gluthathione and gamma-glutamylcysteine
abstract PURPOSE: To fractionate the tilted substances and to recover the tilted high- purity substances form the fractions, by passing a solution containing gluthathione and γ-glutamylcysteine through a weakly basic ion exchange resin and eluting stepwise the solution with acids having different ionic strength. n CONSTITUTION: A solution (e.g., extracted solution from microorganism such as yeast, etc.) containing cysteine, gluthathione and γ-glutamyl-cysteine is passed through a weakly basic anion exchange resin and gluthathione and γ- glutamylcysteine are adsorbed on the resin. Attached cysteine is washed with water and removed. Gluthathione is eluted with an aqueous solution of acetic acid having a concentration to elute only gluthathione and then an acetic acid solution with high concentration, a formic acid solution, a hydrochloric acid solution, sulfuric acid solution, etc., is made to flow to elute γ-glutamylcysteine. The high-purity aimed substances are obtained from the fractions by concentration, crystallization, lyophilization, etc. The ion exchange group of the weakly basic ion exchange resin is a tertiary amine and its ion form is preferably an acetic acid form or a mixed form of the acetic acid form and a free form. n COPYRIGHT: (C)1986,JPO&Japio
