(57) [Summary]nMethods for altering the substrate specificity of plant acyl-ACP thioesterase, and genetically engineered plant acyl-ACP thioesterase produced therefor are provided. As desired for modification for this, two thirds from the C-terminus of the plant thioesterase are identified. Also provided are DNA sequences and constructs for expression of the engineered thioesterase, and novel thioesterases produced therefrom. Such DNA sequences are used for the expression of engineered thioesterase in host cells, in particular in seed cells of seed oil crops for modification of the fatty acid composition. The C12 selective plant acyl-ACP thioesterase described herein can be modified to obtain a plant thioesterase having approximately equal activity on C14 and C12. Further modification of the C12 enzyme results in a thioesterase having greater activity on C14 compared to the C12 substrate. |
