abstract (57) Abstract: The present invention provides a method for detecting a nucleic acid and a target sequence, In particular, it relates to a detection method using fluorescence quenching. A nucleic acid for detection is used for detection of a target sequence of a nucleic acid by a fluorescence quenching system. The nucleic acid for detection comprises at least two oligonucleotides and is partially single-stranded and partially double-stranded. The donor / acceptor dye pair is such that one of the two dyes of the donor / acceptor dye pair is attached to the first oligonucleotide, the other is attached to the second oligonucleotide, and the first and second oligonucleotides are base-paired. When in proximity, donor fluorescence is quenched in close proximity. In the absence of the target, the detection oligonucleotide retains a partially single-stranded and partially double-stranded form. However, if the target is present, the nucleic acid for detection is displaced from the first oligonucleotide and the distance between the donor and acceptor dyes increases, causing a change in fluorescence.
