abstract | (57) Abstract: Active β-galactosidase forms an intramolecular crosslink in the enzyme donor polypeptide fragment of β-galactosidase, thereby forming a cyclic enzyme donor that interferes with complementation with the enzyme acceptor fragment. To form Circular enzyme donors can be linearized by cleavage to restore complementation activity. Disclosed are assays in which such a cyclic enzyme donor is linearized by a particular analyte, and a novel homobifunctional bismaleimide crosslinker of the formula: Where R is hydroxy or acetate. |
