abstract | PURPOSE:To facilitate autonomous propagation using a plasmid pBY503, by synthesizing the plasmid pBY503 having 10 megadaltons molecular weight and the numbers of cleavage sites by restriction enzymes as follows. 4; EcoRI, 1; BamHI, etc. CONSTITUTION:Microbial cells of Brevibacterium.stationis are subjected to treatment with a lysozyme to carry out bacteriolysis. The resultant bacteriolytic solution is then subjected to treatment with phenol chloroform. A plasmid is subsequently recovered from the treated solution into fractions by precipitation with ethanol and isolated by equilibrium density-gradient centrifugation with cesium chloride containing ethidium bromide. The isolated plasmid pBY503 has about 10 megadaltons molecular weight and the numbers of cleavage sites by restriction enzymes as follows. 4; EcoRI, 1; BamHI, 2; SauI, 3; KpnI and 2; SalI. This plasmid is useful as a vector for recombining genes. |
