titleOfInvention Modification reagent for easily measuring hemoglobin derivative in blood
abstract PURPOSE: To measure the ratio of a specific hemoglobin derivative in blood by treating a blood sample with thiocyanate and an oxidizer for modifying total hemoglobin in the sample and determining the specific hemoglobin derivative by immonoassay. CONSTITUTION: A blood sample is treated by a modification reagent containing thiocyanate for modifying all hemoglobin in the sample and an oxidizer for converting the hemoglobin to the form of methemoglobin. Then, the total amount of methemoglobin in the modified blood sample is tested and the amount of hemoglobin derivative in a form being modified by immunoassay, namely hemoglobin Alc, is obtained. A modification process is performed at a room temperature and the concentration of thiocyanate is approximately 3 M or higher. Ferricyanide, chlorate, and nitrite are desirable as the oxidizers. Methemoglobin can easily be measured from the absorbance of light with a wavelength of 540nm. A testing method by particle agglutination suppression is used for immunoassay for detecting the modified hemoglobin Alc.
