abstract |
The present invention provides a method for enhancing the efficiency of introduction of a viral vector into a host cell, such as an unstimulated stem cell. The method includes introducing the vector into a host cell in the presence of a SAMHD1 inhibitor (eg, Vpx protein) and an inhibitor of the mTOR complex (eg, rapamycin or an analog thereof). Kits or pharmaceutical combinations for delivering therapeutic substances into target cells with high targeting frequency and load delivery are also provided in the present invention. The kit or pharmaceutical combination typically contains a viral vector encoding a therapeutic agent, a SAMHD1 inhibitor or polynucleotide encoding a SAMHD1 inhibitor, and an inhibitor of the mTOR complex.
