The invention provides methods for genetically engineering cells, including cells for use in conjunction with genetic engineering. In some embodiments, provided methods comprise transducing cells by incubation with retroviral vector particles, such as lentiviral vectors, wherein prior to incubation, the cells were not incubated with an activator or stimulator, such as without anti-CD3 /anti-CD28 antibody and/or one or more recombinant cytokines were incubated together. In some embodiments, such methods produce features associated with shortening or improving the process of genetically engineering cells. Also provided are resulting cells transduced with a recombinant or heterologous gene, such as one encoding a chimeric receptor (e.g., a chimeric antigen receptor) or other recombinant antigen receptor (e.g., a transgenic receptor) and compositions thereof.
