Some aspects of the invention provide a microorganism that has been engineered for oil production. Methods for microbial engineering and the use of engineered microorganisms are also provided herein. In some embodiments, a combination of steps controlling the rate of lipid synthesis, eg, generating a metabolite for lipid synthesis, acetyl-CoA, ATP or NADPH (push step), and feedback inhibition of lipid synthesis. Microorganisms are provided that are engineered to modulate a combination with a step of sequestering a product or intermediate of a mediating lipid synthesis pathway (pull step). Such push-and-pull engineered microorganisms exhibit significantly enhanced conversion yields and TAG synthesis and storage properties. |
