abstract | A sensitive assay is disclosed that combines immunomagnetic enrichment with multi-parameter flow cytometry or image cytometry to detect, calculate and characterize blood cancer cells. The present invention involves binding different antibodies to the same ferrofluid. This has the effect of making ferrofluid multispecific to the antigen to which it is bound. Multiple antibodies present on the same ferrofluid will not interfere with each other or otherwise interfere. Such ferrofluid has a very favorable effect of being able to specifically bind to two or more types of cells. This assay is particularly useful for enabling capture of CTCs with low EpCAM expression but highly expressing other tumor markers, and as a result, this assay can be used for biological characterization and cancer cell characterization.
