The present invention relates to reducing target gene expression in a cell comprising contacting the cell with an amount of an isolated double stranded nucleic acid (dsNA) effective to reduce target gene expression in the cell. Compositions and methods are provided. The dsNAs of the present invention are deoxyribonucleotide patterns designed to direct the site of Dicer enzyme cleavage within the dsNA molecule (in most embodiments, the pattern comprises at least one deoxyribonucleotide-deoxyribonucleotide base pair).
