abstract To provide a method capable of accurately measuring the pH value of a cell culture solution without depending on the concentration of growth factors such as fetal bovine serum contained in the cell culture solution. In a wavelength region of 300 nm to 800 nm, a first substance (growth factor) having a first absorption peak, a second convergence point and a third absorption peak where the absorbance converges regardless of the second absorption peak or pH, or A medium solution containing a second substance (pH indicator) having a third convergence point at which the absorbance converges regardless of pH is added to the light having the first absorption peak wavelength λ1 and the second absorption peak or second convergence point wavelength λ2. A plurality of lights including a light having a wavelength of λ4, a light having a third absorption peak or a third convergence point wavelength λ3, and a light having a wavelength of λ4 at which the absorbance of either the first substance or the second substance converges regardless of pH. A step of irradiating the medium solution; a step of receiving the irradiated light; and a step of calculating the pH of the medium solution corrected for the influence of the first substance based on the absorbance of each wavelength obtained by receiving the light.
