abstract PROBLEM TO BE SOLVED: To provide a method for enzymatic synthesis of a single-stranded polynucleotide or a double-stranded polynucleotide having a desired chain length. A method for producing a single-stranded polynucleotide having a desired chain length by using polyribonucleotide phosphorylase (PNP) as a substrate and adding an appropriate amount of oligo A to the substrate as a primer. A method for producing a polynucleotide characterized in that the chain length is 200 to 500 base pairs and the amount of oligo A is 1/10 to 1/100 of the concentration of the substrate.
