abstract |
Described herein are methods and compositions for stimulating the growth of cells that express adhesive junctions and cease proliferation, such as human corneal endothelial cells, by downregulating certain cell-cell junctions. In one embodiment, down-regulation is achieved using RNA interference by contacting the cell with an agent that increases mitogenic growth factors and cytoplasmic cAMP. In addition, methods for isolating human corneal endothelial cells from keratocytes and methods for preserving and maintaining the viability of human corneal endothelial cell aggregates are described herein. Also described is a surgical implant comprising human corneal endothelial cells and a biocompatible carrier that have been isolated, optionally stored, and transiently contacted with an agent that downregulates expression of P120. The methods and compositions described herein can be used for novel therapies that help to expand human corneal endothelial cells during in vitro tissue design and for in vivo treatment of corneal endothelial cell dysfunction . |
