Methods and compositions for inhibiting the regulation of T cell costimulatory pathways by pathogens are provided. Inhibition of direct interaction between a superantigen and a specific site within the dimer interface of a CD28 family member using an immunomodulatory peptide, at the dimer interface of a T cell costimulatory pathway member A specific antagonist immunomodulatory peptide comprising an amino acid sequence derived therefrom. The peptide comprises an amino acid sequence that specifically binds to an amino acid sequence within a dimer interface of a T cell costimulatory pathway member, a composition comprising the peptide and a method for the treatment of immune related diseases.
