The present invention relates to the screening of platforms using miniaturized three-dimensional cell chips for high-throughput toxicology screening of test and lead compounds, prodrugs, drugs and P-450 generated drug metabolites. For this purpose, 3D cell chips are aligned on functionalized substrates (eg glass microscope slides) for spatially addressable screening against a large number of test compounds and are as small as 10 nL. Human cells encapsulated in a matrix (eg, collagen or alginate gel) in volume are used. With the platform of the present invention, more than 3,000 cells of matrix islands can be spotted, providing simultaneous screening for multiple compounds in repeated dosing and high replication. |
