titleOfInvention Method for quantifying DNA-binding protein and kit for quantification
abstract PROBLEM TO BE SOLVED: To provide a method capable of quantifying the content of a target DNA binding protein in a liquid sample such as a biological sample containing impurities with high accuracy in a short time without performing separation and purification operation, and a quantification kit therefor provide. In a mixed solution A in which a liquid sample is mixed with a reagent A containing a fluorescently labeled nucleic acid probe Pfw having a nucleic acid sequence to which a target DNA binding protein binds and an unlabeled nucleic acid probe Pw having the nucleic acid sequence. A translational diffusion time Ta; and a translational diffusion time in a mixed solution B in which the liquid sample is mixed with a reagent B containing an unlabeled nucleic acid probe Pn composed of a nucleic acid sequence to which the fluorescently labeled nucleic acid probe Pfw and the DNA-binding protein do not bind. Tb is obtained by fluorescence correlation spectroscopy, and the amount of the target DNA-binding protein in the sample is obtained based on the difference between the translational diffusion times | Ta−Tb |. The quantification kit is a combination of a reagent A containing probes Pfw and Pw and a reagent B containing probes Pfw and Pn. [Selection] Figure 2
