Methods for enhancing substrate properties of terminal phosphate-labeled nucleotides for enzymes, nucleic acid detection methods, and manganese complexes of terminal phosphate-labeled nucleotides. PROBLEM TO BE SOLVED: To describe methods for enhancing their substrate properties for various enzymes using terminal phosphate-labeled nucleotides in the presence of a manganese salt. In particular, methods for detecting nucleic acids in a sample based on the use of terminal phosphate labeled nucleotides as substrates for nucleic acid polymerases in the presence of a manganese salt are described. Further provided are terminal phosphate-labeled nucleotides and manganese complexes of terminal phosphate-labeled nucleotides with enhanced substrate properties and novel linkers. The presence of a manganese salt increases the enzyme substrate property of terminal phosphate-labeled nucleotides, and the method of detecting nucleic acids in a sample using the same and the terminal phosphate-labeled nucleotides having novel linkers therefor are used. Manganese complexes are provided. |
