abstract | The present invention provides methods and compositions for synthesizing oligopeptides or polypeptides by convergent assembly of oligonucleotide pairs in a chemical ligation reaction. An important aspect of the invention is an oligopeptide having a C-terminal disulfide protected carboxythioester group that can be deprotected to spontaneously generate a free C-terminal thioester moiety. This allows a single precursor to participate in a series of chemical ligation reactions, thereby enabling a convergent synthetic approach. The present invention is useful in methods for chemical synthesis of oligopeptides, polypeptides, and proteins, particularly using four or more peptide fragments to assemble oligopeptide, polypeptide, or protein products. In some cases, the efficiency of the natural chemical ligation reaction is improved. |
