abstract | Novel methods for producing recombinant polynucleotides in vitro have been disclosed. The method involves treatment of a heteroduplex DNA sequence with a nuclease (preferably DNase I) and a polymerase (preferably DNA polymerase I), which are enzymes involved in nick translation. The results obtained using this process are superior to those obtained by conventional in vivo recombination tests using a specific DNA repair system. |
