abstract | Nucleic acids encoding engineered DNA polymerases (e.g. an engineered 9°N DNA polymerase) comprising at least one modification in motif A at positions 408, 409 and 410 and/or at least one modification in motif B of the polymerase at positions 484, 485 and 486 are disclosed. More specifically, the claims define an engineered DNA polymerase comprising a variant having at least 95% sequence identity to one of SEQ IDNO:1, 2 or 3 and comprising an amino acid substitution at each of positions Leucine-408, Tyrosine-409 and Proline-410 (L408, Y409, P410). Preferably the substitutions are L408F, Y409F and P410T or L408F, Y409H and P410V. Further modifications D141A and E143A are preferred. Preferably the polymerase is B-type having enhanced base pairing accuracy with a reduced error rate or less than 0.75 per 100 cycles of DNA sequencing. Methods of using the DNA polymerase in cycle sequencing using terminator nucleotides are further disclosed. |
