The invention is directed to a Method for detecting a target moiety in a sample of biological specimens by: na) providing at least one conjugate with the general formula (I) n n A n - P - B m - C q -X o (I) n nwith nA: nantigen recognizing moiety; nP: nenzymatically degradable spacer; nB: nfirst binding moiety nC nsecond binding moiety nX: ndetection moiety; nn, m, q, o nintegers between 1 and 100, nwherein B and C are non-covalently bound to each other and A and B are covalently bound to P nb) labelling the target moiety recognized by the antigen recognizing moiety A with at least one conjugate nc) detecting the labelled target moiety via detecting moiety X nd) cleaving C q -X o by disrupting the non-covalent bond between B m and Cq from the labelled target moiety ne) cleaving the binding moiety B m from the labelled target moiety by enzymatically degrading spacer P. n The method is useful to identify target moieties on the biological specimens. The biological specimens detected by the conjugate can be subsequently removed from the sample. |
