abstract | A method for replacing a p53 gene sequence with an exogenous nucleic acid sequence is described. The method comprises contacting the p53 gene sequence with one or more zinc finger nuclease (ZFN) in the presence of a donor nucleic acid comprising the exogenous nucleic acid sequence. The ZFN comprises a zinc finger peptide (ZFP) portion comprising four or more zinc finger domains, and one or more endonuclease portion. Also disclosed are ZFNs that recognise at least 12bp of a p53 gene sequence and that are capable of cleaving the p53 gene sequence, e.g. to promote homologous recombination and enable replacement of mutated p53 gene sequences. The methods and ZFNs have particularly utility in treating cancers associated with the p53 gene and protein. A method for identifying useful nucleic acid binding polypeptides is also described. The method may be based on a yeast one-hybrid assay. |
