abstract The present inventors have identified the recognition and cleavage domains of the Fok Inrestriction endonuclease. Accordingly, the present invention relates to the DNA segmentsnencoding the recognition and cleavage domains of Fok I restriction endonuclease,nrespectively. The 41 kDa N-terminal fragment constitutes the Fok I recognition domain whilenthe 25 kDa C-terminal fragment constitutes the Fok I cleavage nuclease domain. The presentninvention also relates to hybrid restriction enzymes comprising the nuclease domain of then Fok I restriction endonuclease linked to a recognition domain of another enzyme.nAdditionally, the present invention relates to the construction of two insertion mutants ofn Fok I endonuclease.
